PRILBEN VET 20 mg is indicated for the treatment of heart failure in dogs. The active substance benazepril hydrochloride is a prodrug that is hydrolyzed in vivo to benazeprilat, which inhibits angiotensin converting enzyme (ACE). This prevents conversion of inactive angiotensin I to active angiotensin II, thereby blocking vasoconstriction and sodium and water retention.
Benazeprilat causes prolonged inhibition of plasma ACE in dogs, persisting significantly for 24 hours after a single dose. In dogs with heart failure, it reduces peripheral resistance, blood pressure, and left ventricular load. After oral administration, benazepril is rapidly absorbed from the gastrointestinal tract. Peak benazeprilat concentrations occur at approximately 2 hours, both with and without food. Absolute systemic bioavailability is about 9%.
Steady state is reached in less than 4 days. After a single dose of 0.23 mg benazepril per kg body weight, peak benazeprilat concentration is 40.9 ng/ml at 1.5 hours, with an AUC of 320.5 ng/ml/h and a half life of 12.4 hours. The drug is eliminated equally via hepatic and urinary routes.
Each tablet contains 4.6 mg benazepril (equivalent to 5 mg benazepril hydrochloride). Excipients include titanium dioxide (E171), yellow iron oxide (E172), red iron oxide (E172), and black iron oxide (E172).
