CLAVASEPTIN

Laboratory
VETOQUINOL EVSA
Pharmaceutical form
Oral tablet (Comp.o.)
Composition by tablet:
Amoxicillin (in the form of trihydrate) 200 mg; clavulanic acid (as potassium salt) 50 mg; brown iron oxide (E-172) 0,475 mg.
Pharmacological properties
Pharmacodynamic properties :
• Amoxicillin is an aminobenzylpenicillin from the â-lactam family that prevents the formation of the bacterial cell wall by interfering with the final stage of peptidoglycan synthesis.
• Clavulanic acid is an irreversible inhibitor of intracellular and extracellular â-lactamases, protecting amoxicillin from the inactivation produced by them.
• Amoxicillin/clavulanate have a broad antibacterial spectrum including both Gram positive and Gram negative aerobic, facultative anaerobic and strict anaerobic â-lactamase producing strains.
Amoxicillin/clavulanic acid breakpoints (NCCLS/2002):
staphylococci: Sensitive: MIC<4/2 µg/ml, resistant: MIC>8/4 µg/ml.
Other organisms: Sensitive: MIC <8/4 µg/ml, resistant: MIC >32/16 µg/ml.
The combination amoxicillin/clavulanic acid in a 2/1 ratio has shown the following sensitivity data in periodontal infections in dogs in Europe (isolated in 2002 in France, Germany and Belgium):
– Streptococcus spp.: MIC90: 0,4/0,2 µg/ml.
– Pasteurellaceae: MIC 90: 0,4/0,2 µg/ml.
Escherichia coli: MIC90: 5,3/2,6 µg/ml.
– Enterobacteriaceae: MIC 90: 22,6/11,3 µg/ml, except for Enterobacterr spp.
– Pseudomonadaceae: MIC 90: 147,0/73,5 µg/ml.
– Enterobacter spp.: MIC90: 49,6/24,8 µg/ml.
• Resistance to â-lactam antibiotics is mainly mediated by â-lactamases, which hydrolyze antibiotics such as amoxicillin. Resistances appear mainly in Pseudomonadaceae (81,25%) and in Enterobacter spp. (55,5%).
Pharmacokinetic properties:
Following oral administration at the recommended dose in dogs, the absorption of amoxicillin and clavulanic acid is rapid. The maximum plasma concentration of amoxicillin is 8,5 µg/ml and is reached in 1,4 h and the maximum plasma concentration of clavulanic acid is 0,9 µg/ml and is reached in 0,9 h.
Removal is also fast.
After repeated administration of recommended doses, no accumulation of amoxicillin or clavulanic acid occurs and steady state is reached rapidly after the first administration.
Interactions and incompatibilities
The bactericidal activity of amoxicillin can be reduced by the simultaneous use of bacteriostatic substances such as macrolides, tetracyclines, sulfonamides and chloramphenicol.
Indications and target species
Dogs: Treatment or adjunctive treatment of periodontal infections caused by the following bacteria sensitive to amoxicillin in combination with clavulanic acid, for example, pasteurella spp., Streptococcus spp. and Escherichia coli.
Contraindications
• Do not use in case of hypersensitivity to penicillins or other substances belonging to the âlactam group.
• Do not administer to gerbils, guinea pigs, hamsters and rabbits and use with caution in other small herbivores.
efectos secundarios
• Vomiting and diarrhea may appear. Treatment may be interrupted depending on the severity of the observed adverse effects and the benefit/risk assessment performed by the veterinarian.
• Hypersensitivity reactions (allergic skin reactions, anaphylaxis) may be observed. In these cases, administration should be discontinued and symptomatic treatment initiated.
Route of administration
Orally.
Posology
The recommended dose of the product is 10 mg amoxicillin/2,5 mg clavulanic acid per kg twice daily orally in dogs, or 1 tablet/20 kg/12 h according to the following table:

Liveweight
(kg)
Tablets to be administered
2 times a day
8,1-10 1/2
10,1-20 1
20,1-30 1 1/2
30,1-40 2

Duration of treatment: 7 days.
To ensure correct dosing, the animal's weight must be determined as accurately as possible.
possible to avoid underdosing
overdose: At doses three times higher than the recommended ones administered during 28 days,
diarrhea was observed in dogs. In the case of overdose, symptomatic treatment should be initiated.
